Zoledronic acid: clinical potential beyond the prevention of skeletal complications in patients with cancer

10.2217/14750708.4.3.241 © 2 Zoledronic acid has received international approval for the treatment of bone metastases and bone lesions from cancer. Phase III studies established the efficacy and safety profile of zoledronic acid in a broad range of tumor types. A direct comparison study with pamidronate demonstrated that zoledronic acid had a similar safety profile and provided superior clinical benefits in patients with breast cancer. Exploratory analyses of the large Phase III databases have demonstrated a survival benefit in patients with breast or lung cancer who achieved normalization of their biochemical marker of bone resorption, N-terminal telopeptide of Type I collagen, within 3 months of zoledronic acid therapy. Ongoing studies are evaluating zoledronic acid for the prevention of accelerated bone loss associated with aromatase inhibitors in patients with breast cancer and for the prevention of new bone metastases.

Zoledronic acid in the management of metastatic bone disease
Many patients with advanced cancer experience decreased bone strength due to metastatic foci, underlying osteoporosis and/or cancer treatment induced bone loss. The clinical consequences of metastatic disease involving the skeleton are widespread. This review focuses on the efficacy, pharmacology, and safety when using intravenous biphosphonate such a zoledronic acid for cancer bone metastases....
متن کاملTreatment of bone metastases and bone pain with bisphosphonates.
Many solid tumors metastasize to bone, leading to debilitating skeletal complications such as intractable bone pain and pathologic fractures. Patients who experience a skeletal-related event (SRE) are at higher risk for subsequent events. After an SRE such as a pathologic fracture, spinal cord compression, or the requirement for orthopedic surgery or palliative radiation therapy, a patient's qu...
متن کاملLong-term efficacy of zoledronic acid for the prevention of skeletal complications in patients with metastatic hormone-refractory prostate cancer.
In a placebo-controlled randomized clinical trial, zoledronic acid (4 mg via a 15-minute infusion every 3 weeks for 15 months) reduced the incidence of skeletal-related events (SREs) in men with hormone-refractory metastatic prostate cancer. Among 122 patients who completed a total of 24 months on study, fewer patients in the 4-mg zoledronic acid group than in the placebo group had at least one...
متن کاملNew Research Findings on Clinical Benefits of Bisphosphonates in Patients With Advanced Prostate Cancer
Objectives: Bone metastases develop in the majority of patients with advanced prostate cancer, and this fact puts these patients at high risk for potentially debilitating skeletal complications. Emerging therapies for the prevention of these complications are important for preserving patients’ quality of life. Methods: Existing and novel therapies were identified and researched through PubMed a...
متن کاملEconomic evaluation of zoledronic acid versus pamidronate for the prevention of skeletal-related events in metastatic breast cancer and multiple myeloma.
Skeletal complications of cancer decrease health-related quality of life. Bisphosphonates can prevent skeletal-related events. We collected resource use data prospectively for 930 patients alongside a multinational trial of zoledronic acid versus pamidronate for patients with metastatic multiple myeloma or breast cancer and > or =1 bone lesion. Country-specific unit costs were assigned to count...
